Alginate-Methacrylate Photoresins: Source, M/G Composition, Methacrylation Route, Covalent-Ionic Dual Networks, Hydration, Degradation and Qualification

Primary studies show that photocrosslinkable alginates are route-specific macromers, not interchangeable grades. Carbodiimide coupling of 2-aminoethyl methacrylate enabled substitution-dependent swelling, modulus and degradation; glycidyl methacrylate, 2-aminoethyl methacrylate and methacrylic anhydride produced derivatives with similar measured substitution but different compressive failure behaviour; functionalisation time and initiator concentration changed a separate photocurable bioink system; oxidation before methacrylation created another degradation coordinate; and light-patterned ionic alginate demonstrated that divalent-cation-bound networks can be fabricated without covalent methacrylate chemistry. Qualification must therefore freeze alginate source, molecular-weight distribution, M/G composition, modification route, measured substitution, oxidation, purification, concentration, ions, initiator, absorber, dose, working curve, conversion, wash, hydration, dimensions, mechanics, degradation protocol and model-specific biology. Calcium treatment is not proof of a covalent dual network, exposure time is not conversion, and one degradation profile is not a universal resorption rate.
